I posted a detailed reaction in my personal journal and I thought it'd be fun to post here too, for a discussion with all my lovely fellow fans :).
- Dean = Jesus Christ Superstar versus Sam = Anti-Christ. Glorious angst much!
- I can't see Sam and Dean fighting each other, even if god and devil tell them to. Humans have free will hence they are not the soldiers of a god nor are they sons of a devil. And I don't think either brother prioritizes anybody over each other.
- It would be delicious if the sides and players of Heaven and Hell are not so neatly aligned; namely, rogue angels and rogue demons. Ruby for example. She claims to be against Lilith but is also apparently terrified of Castiel so where does she stand?
- Lilith has to break 66 seals to free Lucifer. Why and how and by whom is Lucifer locked up in the first place? And are Lilith and Lucifer going to be the dark side version of Adam and Eve if she succeeds?
- What if Castiel is a vigilante angel? Maybe god has not commanded angels to fight but a group of angels have taken it upon themselves to do so anyway?
- I love how conflictingly binary and on-binary Dean and Sam's beliefs are when it comes to angels, demons, god, the devil, evil, good, divinity, humanity, sin and redemption. God created the devil so if you don't believe in the devil or god, then you don't believe in the other? If you believe in hell, do you believe in heaven? If you don't believe in redemption, do you believe you're damned because you're a hunter who has saved many but also not saved some? You don't believe in heaven but you believe in hell so how does that square with your belief that you're not worth redemption and hence neither peace or heaven but your brother is, no matter what he does and you will give everything to keep him from hell but what happens to his soul, then, when he dies?
- Sam is quick to believe in angels and god. Where does the devil come into it given that a demon is teaching him about the dark side and Casey told Dean that the devil is as god to demons. Of course, if god created all of them then maybe Sam is on a path to god too?
- What is Sam's identity then? Human? Half-demon, half -human? He didn't choose to have Azazel's blood so does it count in the final battle? And what was Azazel's plan for Sam given that Lilith wants to bring about the apocalypse and free Lucifer and rule all yet she is against Sam? Was Azazel a challenger to Lilith yet with the same plan? Or was he a rogue demon working for god against Lilith and Lucifer and that plan went bust so god sent down Castiel?
- And by the way, who says Sam is the Anti-Christ and will stand against god and with the devil? Especially since he's against Lilith and Lilith wants to free Lucifer presumably so Lucifer helps her? In which case - who's side is Lucifer going to be on, Sam's or Lilith's?

I'm going to shut up now. Heh heh.
