Personality:
The first thing you'd notice about Derek if you met him, was his kindness and niceness. Even to strangers, he wants to help them, to get over whatever happens and to help them smile again. In a paraphrased version of his own words, 'a doctors work isn't just on the operating table, our bed-side manner is also important'. His smiles barely wavers, as long as other people need his help. When he notices a pretty lady, however, he'll be a little awkward, and might try some light flirting.
He's also determined, with everything he does, and is completely focused when operating on somebody.
However, with all of this good, he also has a little bad. How, you might ask? He's lazy, for one. He'll put off non-operation work for ages, and is still sometimes getting to work late, and sleeping in instead. This has almost stopped with the help of Angie, but it's still slightly noticeable.
He also has an overconfidence streak. This comes from the sudden reputation he's gained, as the savior of the world from GUILT. So it's a little justified, but he will talk it up a bit.
Also... when operating, he takes the responsibility of the patient up all by himself. If he fails, he takes it personally, and will quit doctoring forever. Luckliy, he is pre-UTK2, so he hasn't canon-ly lost a patient.
At the canon point I've taken him from, he is infected with GUILT, and it will slightly affect him. One of the sorts he is infected with, Kyriaki, is shown to release certain chemicals if not treated, and those chemicals cause depression. However, that won't affect him much in the beginning -- but it will amplify his feeling of lost and homesickness, if only a little. (The other sort is Paraskevi, but that doesn't affect personality at all. In fact, it's just meant to kill him. :p)
As an aside, it's shown in Under the Knife 2 that he is slightly oblivious to love, when he is with Angie at least.

Strengths/Weaknesses:
+ Derek has the powers of the Healing Touch, which in canon, allowed him to focus and slow down time. It only works with good doctors...
+ Which means that Derek is skilled as a doctor, anyway. He can make miracles happen, and save people who would have died afterwards.
+ Even without special powers, he can focus on something well, and almost get to a point of obsession with it...

- ...but that can mean he gets occupied and forgets to do other things.
- He is not good with puzzles, and struggles with them.
- He suffers from laziness problems, and needs to be disciplined by Angie or somebody similar.
- He may jump to incorrect conclusions -- not as much as he used to, but still.

First Person Sample: [Open to a very dazed looking doctor, stumbling out of 'mom's house. Who's holding his gear? It's his starter, by his side.]

--ler! This isn't funny, you know, you can get out of that outfit now! And this isn't proper recovery protocol... am I even in Europe anymore? The hills look more like somewhere in Japan...  or what Dr. Weaver told me about it, anyway.

[He then looks straight into the camera.]

Uh... if anybody out there knows how to get to Caduceus Europe, could you let me know? There's a meeting soon, and I've recovered enough to go. It'd be bad if I didn't end up going, I'm the guest of honour.

Or, at least, does anybody have a useful map? This one in this bag isn't that good...

Third Person Sample: The cold chill of a winters night was, luckily, something that Derek had never experienced. Well, at least, until tonight. He sat under a tree, looking out into the distance. "It's nearly time to sleep, isn't it? Angie's gonna be mad if she finds out it's nearly midnight, and I'm still awake," Derek said, trying to hold back his yawn. But he couldn't sleep yet -- he had to find his glasses, or at least help his round, pink monster help. If he had lost those... well, it would be hard to get a replacement. Unlike on Earth, he didn't have any medical history here, and he'd have to go for another eye exam. Did they even sell his type of glasses here? Derek let out another yawn, and rubbed his eyes. He was so tired, and those glasses were... well, who knows where he dropped them. However, he heard flapping behind him, and then felt a hard hit on his head. It was a wild Pokemon, but Derek could only see those red eyes. "Tyler...?" he called, ducking. His Chansey ran to his side, and quickly shielded him from the wild Hoothoot. "Tyler, use Defense Curl! I'll try and get us away!" Derek said, turning away. The Chansey, however, used Pound instead. No way was it going to let Derek get hurt again. As Derek fled, Chansey kept on attacking the Hoothoot, and managed to make it faint. By this time, Chansey was also quite tired. Derek turned his head, and then his glasses slid onto his face. They'd been on his head the whole time. "Tyler...!" he said, crouching down to the fainted owl. "We'd better get it to a Pokemon Center... I can't believe you'd hurt somebody! That's not in your nature." he said, returning Chansey to his ball. It would be too dangerous to just carry the young owl, Derek thought, as he took out another Pokeball, and captured the weakened beast. Of course, he'd release it after it was better.
