o.O You put a female in the same cage with two males? And they haven't killed each other? That's usually not a good idea. Males are VERY territorial, especially when it comes to females. They HAVE been known to kill or at least severely injury other males because of females (and not even because of females - most males simply cannot share a cage with another male; they usually end up living alone).
I had one male mouse with a HUGE lump on his testicles. It was definitely a tumor. It's quite possible this is what your mouse has if it is in addition to his testicles. But you may want to check and make sure it isn't a wound.
