So, Dean is the moral compass with all things human, keeping his brother and father from going off the rails. Sam is the moral compass with all things supernatural, teaching Dean to see the world differently.
I'm glad that the mythology does get resolved. I hate shows that contradict themselves and drop hints that go nowhere. Is Jo Ellen's daughter? So far, I see no reason for hating on her at all. She comes off as a little cocky, but I think that's normal for the young. And I"m in love with Ellen. I can see Ellen really domming Dean. He needs someone to give him permission to let go.
Jo is Ellen's (and, sorry, I forget her husband's name) daughter. That becomes clear later on in season 2.
I think the reason people hate Jo is because she might be an obstacle between the boys: either because of a love interest interfering w/ an intense familial relationship or the whole wincest phen.
A couple of other comments. Clown. You said that you see Dean pulling away from his father's beliefs. What in this episode lead you to this? I see this in him questioning and standing up to his father in the last few epis. in season 1 and other points that you probably haven't seen yet.
Bloodlust. I agree w/ you that Sam probably gave Lenore blood. There are some fanfics about this. Though, what I remember about this episode is Sam saying that only Dean gets to call him Sammy, however you want that to mean.

I don't think John trusted anyone enough to be a real part of the community.
