Today I'm sporulating a/alpha spo71-myc-trp as well as my WT (again). It seems the last 2 times I sporulated WT they were contaminated. I must be more aware of how I am handling my samples. I have also begun looking at my samples under 40x to detect any bacteria as soon as possible. So far I do not have any rod contamination, but my CB79 (myc strain above) may have cocci. I've never seen it before so I must keep checking to see if it doubles significantly.
We received a new imaging system at work, which seems to be a bit better than film, and is (finally) allowing me to develop my gels longer.
Thursday I looked at my CB103 and CB 104 strains (ZZ tagged). Something a bit strange seems to be occurring. There are bands in each of the tagged lanes, and not in the WT (this is a good thing), but the bands are not at the predicted size (should be about 175kD), we are seeing bands at about 145kD. My advisor seems to think the predicted size of our protein is incorrect, and wants me to predict a new size (in frame of course).
I will compare my myc tagged band to a new predicted protein size and see what’s what.
