The only thing I can think of that makes even Glee-sense is that this is part of the Blaine/Sue feud and is either some kind of side bet on their feud. I'd figure that Sue is mean to the Cheerios, maybe Tina in particular, and Blaine challenges her about her methods. She probably will take offense and remind him that SHE is the coach, not him. They probably will decide to settle it with a sing-off (because why not I suppose, it's no stupider than anything else), with the terms being if Sue loses, she stops doing whatever thing it is that sets Blaine off; if Blaine loses, he has to hire a plane to fly a banner saying this phrase and not question her authority again.

Of course if Sue hires the plane it would be clear harassment, obviously it's a reference to being gay and to sexual practices, both of which are totally inappropriate comment for a teacher, but that has never stopped her before (though I'm not sure that Sue has been that public about it before). I saw the comment above about Sue drawing attention to Blaine being a "bottom" might cause him to be harassed or threatened in school. This is where Glee sometimes gets a bit erratic, tbh. Sue has always been inappropriate, sometimes even physically dangerous and assaultive even toward her own students. Other times, she claims to be aware of certain dangers and want to protect her students from those dangers (Unique, Kurt, Santana). Having her publicly shame Blaine for his sexuality seems to be a new low for her. However, the way the show seems to be packaging Blaine is that he is popular/successful or at least not a target even though it is known that he is gay.

I think that if Sue trounces Blaine in this sing-off it is probably part of what Darren was talking about as far as making Blaine struggle a bit more, and not having him win everything without really even trying or caring about winning. Now that things are looking more optimistic than they were with Kurt, this might be the show's way of taking Blaine down a few pegs so the audience can semi-relate to him. Losing the Diva-off to Tina might be part of that and I have a sneaking suspicion something is going to screw up his admission to NYADA so that if Blaine goes to NYC at all, it will be in some less triumphant way than as a NYADA freshman. But we'll see.
