Would it just be a walk-in store, or would you do internet sale as well? Anime Nation I know is one that does huge sales online, but they also have a walk-in retail store at their warehouse ... albeit not a very extensive one; it felt more like walking into Suncoast what with all of the domestic merchandise. ... Or you could tour conventions as a dealer, too.
I know I'd be looking for things like doujinshi, or the kinds of random trinkets that you find on animate-usa that I usually just refer to as "stuff" because there doesn't seem to be any other category. Keychains and stickers and pins and random whatever, as long as it has my favorite characters on it I'm good. ^_^;; As far as genres, well, I'm a sad, sad PoT fangirl right now so it's about all I buy, unless it's something new or hard to find (ie Last Exile).
I think it'd be cool if the name didn't have "Anime" in it -- too hard to distinguish from all of the other places, you know? But something to set it apart ... and yeah, maybe brand-able. Because then you could get into things like t-shirts and bags and other items unique to your own store.
... and cosplay items! That would be cool.
Promotions ... well. Maybe websites like cosplaylab.com or cosplay.com, or you could maybe get in touch with some of the anime-related fan forums? (I have contacts at animemetro.com, even if I don't use the forums there, personally.) Flyers at conventions are always good, too. And link exchanges, possibly? (Heck, if I ever got my site up and running I'd be happy to exchange, but, ah, as it's currently nonexistent that doesn't do you much good. >>; )
