I just got CS5 myself and I have had a handful of weird glitches, always in regards to some other program (like Windows7 Flicks) but I haven't had a problem with saving anything. Is the option even there under File? Is it greyed out? Can you still use the keyboard shortcuts without exiting?

Ctrl+S = save
Shift+Ctrl+S = save as
Alt+Shift+Ctrl+S = save for web and devices

The option isn't grayed out, just when I click on Save nothing happens, and the keyboard shortcuts don't work either. :/

have you checked adobe's website for fixes that might be available?

I actually did, I don't know whether I'm wording the problem wrong or something like that, but I can't find it.

the easiest way to do it is from within photoshop, from the menu bar, go 'help>updates...' and let photoshop update itself. it will install any/all fixes/patches that have been released. if you haven't done it, give that a try and then restart PS and see if it fixes your issue.

You mentioned trying 'save' and 'save as', have you tried 'save for web & devices'?

Yeah, I remember being confused when things wouldn't save when I tried CS5. Until I tried the new-fangled option. ;)
