I'm not sure how Mary would tie into Ava, but there is that statue of Mary in the church attic where Anna is hiding, and in the Bible Mary's mother was named Anna. Plus there's always Mary Winchester. :)
ETA - Not only are roses associated with secrets through sub rosa, it turns out the blue rose in particular is associated with secrets. "Blue roses traditionally signify mystery or attaining the impossible. They are believed to be able to grant the owner youth or grant wishes." http://en.wikipedia.org/wiki/Blue_rose
So I definitely don't think it's a coincidence that Sam spilled his guts in the Blue Rose motel. :)
