Antonio's not quite the most developed character in Trinity Blood, we're given little details about his personal life; for what I can tell he has no dark and tragic background (TM). He hardly introspects, so don't expect an EMO POST unless something big happens. I've compiled a list of questions that I've taken in account both canon information and my own filling the gaps that helps me to play him. If you have a doubt, ask away!

Name: Antonio de Borgia y Borgia.
Ranks/Titles: Cardinal, Duke of Valencia, son/heir of the Prime Minister of the Hispania Kingdom (Lord Carlos de Borgia), head of the Ministry of Public Affairs in the Vatican. He's the Vatican's PR. TokyoPop claims he's the Prince of Hispania. Let's roll with that.
Powers: Genius. Top student at Cologne University with seven doctorates by the age of twenty-three.
Age: 25 years old.
Appearance: "He was an impressively handsome young man, with heavy eyelids and long hair that cut a rather undignified appearance for a church worker." ROM II. Antonio has dark blond hair and green eyes.
Sexual Orientation: Flaming heterosexual.

✗ What are his thoughts on the City?
Antonio is v. confused about this place, the magic, the danger, etc. But he doesn't allow others to see this or himself to realize this. He's a big pile of denial. Instead, he takes it as an adventure, Antonio's bold when he feels safe and he feels safe with Abel Nightroad around.
✗ What are his thoughts on Abel?
Despite his opportunistic nature, I think Antonio values Abel as a friend and ally. Abel saved his life twice (bullet shield >>) and he's willing to give him a hand to help him in Imitation Star to rescue Esther when even Caterina didn't. He does enjoy poking fun and teasing him, but I think is sort of fraternal affection from an annoying little brother to a silly big brother. So he's going to stick with Abel when he needs him and expects Abel to help him the same. It's probably the lack of formal respect for ranks Antonio truly has that doesn't really see him as a subordinate.

Furniture. Unobstructive. Reliable. Useful.
✗ What are his thoughts on Caterina?
Honeeeeeeeeey. Antonio's half serious about his advances to her. I don't think he's in love with Caterina, but he's attracted to her and there is a genuine concern because he's not a sociopath and actually cares about people. She's probably the woman he's willing to marry for the political alliance and the looks don't hurt, either.
✗ What are his thoughts on religion?
I regard Antonio as an Agnostic. Not quite faithful, not quite atheist (like Abel), just a middle ground, enough to take advantage for this for his increasing power.
✗ What are his thoughts on the Inquisition?
Double edged sword. They follow Francesco who, like Caterina, is a political rival/ally (though, Antonio "likes" both of them). He seems fond of Petros and doesn't trust the others.
✗ What are his thoughts on vampires?
First of, Antonio isn't from the Vatican Domains, he was born and raised in Hispania, plus his education was continued in Germanicus. He'll be more tolerant with vampires than those who hail from Roma or had a tragic past with vampires (like Hugue). In fact, it's likely some of his family may be Methuselah. It isn't uncommon the existence of several vampire nobles in Terran countries Courts. Borgia keep their business to themselves and I doubt they would be questioned by the Vatican so easily. However, there is a difference between a vampire from Hispania, Albion, Franc or Germanicus and one from the Empire. It's not much about species but enemy nation and a danger for his power. So he'll be leery about that. ;)
