Let's set aside the absolutely hilarious face of the lady and focus of Sam and Dean's hands. Not just that they're holding hands but the WAY they are holding hands. Click-click-click to make it humongous and then zoom in on their hands. (How many times can I say the word 'hands' in one post? Apparently endlessly. Handshandshandshandshands.... *g*)
If that doesn't scream for a "Yes, it's true" icon or possibly, "And that's when Jensen said yes..." I don't know what does!
