Yes, keeping the three of them together is his priority because that's all he has, so he takes pot shots at the "normal" life Sam wants because it threatens the only kind of family life Dean believes he can have. But, in his dreams, he'd rather have the typical "apple pie life" with Mom, Dad and Sam, but he knows it's a pipe dream. (That's the deeper meaning behind the running gag that Dean always wants pie, but never gets it. SPN has an interesting relationship with humour. So often superficially lighthearted gags or scenes actually have something sad or dark lurking beneath them.)
Same with Dean's teasing of Sam. It seems light-hearted, but he's actually unconsciously revealing his own insecurities. That's psychologically accurate to real life. When people mock others, it typically comes from a place of personal insecurity and says more about them than the person they're mocking.
