Places you really need to hit...hmmm...The Japanese Tea Garden in GG park is a must, but go early before the tourists get there (or on a rainy day). Find a taqueria in the Mission that looks like it might fail it's next health inspection and have a mind-bogglingly good burrito. The BiRite Creamery at 18th and Dolores has incredible ice cream (all organic, with vegan options). And there's a really awesome Burmese place at California and 5th ave if you get the chance.
As far as the Polk, how the fuck should I know? I'm a Castro girl :-) Pick up the local gay rag and find yourself a bar that looks intriguing.
My favorite Castro scene was the young girl scouts who had set up at 18th and Castro. They had a table, and they'd arranged the boxes in stacks, red orange yellow green blue violet. They were selling like fucking mad - and I just thought - wow, what a cool city. I've realized that it's not the gay people who I love in SF - the gay people there are basically just your garden variety homos, with the same problems and attitude and bullshit you get everywhere. What's awesome about SF is the straight people. They have the best straight people in the whole world.

I'll check it all out. At exactly this moment, actually, I'm at a friend's apartment just above the Sausage Factory. The sign glows just outside the living room window. It's nice that there are some Faeries here.
It's taken me a while to adjust to the whole 'city' thing, but now I really DO like it here. The straight people here really are pretty great.
