There are no Ancients on the ship anymore, the Destiny is on a pre-programmed course to connect the whole Stargate network to the far reaches of the galaxy [or something like that]. The Ancients were supposed to use the ninth chevron to reach it, but they ascended instead and the ship kept going. This is from the official announcement for the show:
After unlocking the mystery of the Stargate's ninth chevron, a team of explorers travels to an unmanned starship called the Destiny, launched by the Ancients at the height of their civilization as a grand experiment set in motion, but never completed.
What starts as a simple reconnaissance turns into a never ending mission, as the Stargate Universe crew who discovers the ship is unable to return to Earth, and they must now fend for themselves aboard the Destiny.
The crew will travel to the far reaches of the universe, connecting with each of the previously launched Stargates, thus fulfilling the Destiny's original mission. Challenges will arise though as the ship comes into range of Stargates placed centuries ahead of the Destiny and the crew is unable to control the ship's navigational schedule. If someone is left behind, there is no way to go back for them, adding to the drama of encountering new races, enemies and adventures.
re: Dr Rush, I'm still not sure he's actually the lead of the expedition, because the casting sides for Colonel Young had him the expedition leader -- and the sides clearly said they wanted someone with a "star name" for that part as well. [Dr Rush didn't have any casting sides out for him, my guess is they wrote it for Carlyle?]
Heh. I'm not sure how much of that comes from the actual TPTB and how much is PR spin from the MGM/Sci-Fi Channel people. Still, it goes with the "don't leave anyone behind OR ELSE" vibe Brad Wright likes so much.
