Buy Grand empties unless you can find Grands with Grand souls. Use anything less for small value stuff like nightvision, water walking, water breathing, etc. Use the rest of them to recharge your items. The smaller ones are nearly useless for enchanting, unless you want a five point fireball.
Also, don't forget that you can use the Oblivion stones you got to enchant weapons. They level scale, so the ones you picked up later will be more powerful.
Personally, I like to put 1 sec soultrap on a weapon, so that they're always soultrapped when I hit them the last time. I also recommend making rings with water breathing, walking, nightvision, etc. On/Off powers can be put on rings and whatnot using small gems, and having a set of rings on you doesn't weigh much.
A bunch of it's just trial and error. Sometimes it makes more sense to do a 2 second 25 point blast than a 1 second 50 point one, because the power of your soul gem not only determines how powerful an attack you can enchant, but also determines the number of charges. Nothing worse than building a sword with +75 fire, only to discover it has four charges.
Finally, the soul gems in the various magic guilds respawn after a few ingame days. I got most of my early Grands and whatnot from my own guilds. for filling the big boys, I find that Fort Nickel, with its penned in denizens do a great job filling really big gems.
