Hie thee to the closest 24 hour pharmacy (the CVS, formerly SavOn, on the corner of Verdugo and Hollywood Way) and buy the highest dose lysine you can find. Take two of those tonight and one per day until the canker sore is gone. The stuff really works. And in the future, take the lysine at the first sign of a canker sore and it'll go away almost immediately.
