There is so much to be said about John that Sherlock could probably write a book on him. He's not as intelligent as Sherlock, but he respects him (in his own Sherlock style, at least); his soldier's fortitude and loyalty make him invaluable as an ally. He is the only person in the world-- any world-- Sherlock would even think of considering as a friend. He forces Sherlock to confront the fact that he does have positive emotions for people, especially when the Core brought him back unexpectedly. If any of his enemies in the Port were to try and hurt John, they can expect a fearful amount of merciless retribution. At the same time, Sherlock knows that John has his back at any given time and would do the same.
UPDATE: Since John left and returned, the two have been through a lot together. The bounty on Batman, John getting drugged, Sherlock dealing poorly with Carrie's death-- it's only made it clearer that this friendship is a binding, beneficial thing for the both of them. They're willing to kill for each other and die for each other. For Sherlock, this is the greatest expression of loyalty.

Sherlock considers plenty of people his enemy, even his own fairly innocuous brother, but he never means the term more when he uses it to describe James Moriarty. The consulting criminal is the yin to Sherlock's yang and one of his few equals in the world. The difference is that Jim is a killer and Sherlock is a savior. He's forced to acknowledge this when Moriarty terrorizes London with a series of puzzle-oriented bombings, and finally, when he takes John hostage. This is the man he could have been if one wrong turn led him to psychopathy. Sherlock superficially hates a lot of things, but there's no one he really hates more than Moriarty. Gone. Good.
quackery (Princess Tutu) - ♙
A young girl caught up in the Heart Theft incident. Seems to be a rock of hope for a lot of people on the NV for some reason, but Sherlock is somewhat indifferent to that. Another gullible one who may be useful for information if he ever needs it. Also a duck
remembered (Doctor Who) - ☆ ♘
A ginger, Scottish pest. Were he in London, she would be of no particular interest at all, but he couldn't help but notice a countryman in the Port. Then came John's unfortunate decision to let her live with them for a few weeks when her first month lease ran out. Laundry was mixed, his things were moved, awkward bathroom sharing was had. Sherlock didn't like it, and he doesn't really like her. Especially if she's one of John's distractions.
aslandish (Chronicles of Narnia) - ★★ ♕
Sherlock has found himself strangely comforted by the presence of Aslan in Siren's Port, though unwillingly. Interacting with a fictional character he read about in his childhood briefly is somehow more unnerving than aliens, demons, mutants, or superheroes. Probably because of what comes with Aslan's symbolism. Still, he can't deny how fascinating it is.
kingofrooks (DC Comics) - ♕/♗
As of now, Sherlock doesn't know that Bruce and Batman are one in the same. He has suspicions, as the last time Bruce Wayne was in the Port, a few incidents that resemble the Bat's MO occurred in passing. Not to mention a noted absence of Robin around. Other evidence gained from interacting with Batman himself lend to Sherlock's suspicions, but he doesn't have enough data to prove anything.
That aside, Sherlock's relationship with Batman has been somewhat turbulent. Though they clearly have similar goals, their different motivations and methods cause them to clash. Sherlock finds Batman's way overly theatrical and ostentatious. He feels this way about any caped heroes that have appeared in the Port, but none have ever explicitly interfered with his own cases. Sherlock has managed to hold his ground so far, but something, whether it be their mutual contacts in both Robin and Re-l Mayer or mutual enemies in the Rogues, will more than likely force them to work together in the future. He won't admit that.
UPDATE: Sherlock was able to deduce and prove that Bruce is Batman, but he doesn't really care either way, so it's a rather apathetically kept secret. Though he respects Batman's skill, Sherlock really resents him in some ways. Most recently and important of all was Carrie's death. Bruce was supposed to be in charge of her; he inspired her to join in all this mad hero business. He could keep tabs on a million different things in the city at once, but he couldn't save the fifteen year old girl he was in charge of. Sherlock knows it's probably not entirely right to blame Bruce, but he does anyway. Whether he'll forgive him or not is up to debate.
girlyboywonder (DC Comics) - ★★★ ♖
Admittedly, Sherlock would have ignored her after their first encounter. That is, if she ever left him alone. Unfortunately, Robin stuck to him like glue, in her ridiculous outfit and all. Because of her flashy methodology and how impressionable she is, he's decided to try and show her how much better his ways of investigation are. So, accidentally, and in a way he'd never admit, she's something of a student to him. Sort of. He just wants to point out how stupid costumes are. And, in some ways, she reminds him of himself when he started down the road of deduction. A female, far more chipper him, anyway. But she actually got a Christmas present from him. Le gasp. They continue to have awkward moments of almost familial affection to each other. Sherlock was inspired to involve himself in the Heartless incident after Carrie, without a heart, killed someone right in front of him. Not particularly phasing, but he knew it was something she wouldn't take well upon her return to normal. Later, when John was taken back home, she tried to console him, key word being tried. He didn't really understand the concept, but now they seem to have a fairly regular father daughter tea time. But he still thinks her costume is stupid.
UPDATE: When Sylar killed her, Sherlock was the closest to his breaking point in the whole year he'd been at Siren's Port. Someone he actually cared about was murdered, the killer was dead, and there was nothing he could do. It forced him to acknowledge that he'd grown to give a damn in some part about the people in this world other than John, and to examine the personal repercussions of the crimes he investigates. He didn't like it. When she came back, the happiness was almost even more baffling.
She seems to have gone home, and while he might admittedly miss her, at least she can't get killed by anything in the Port anymore. Hopefully.

A rather clueless girl from an alternate past. She managed to get herself mixed up with Sionis, and worked at his restaurant without realizing who owns it. Obviously nothing in the way of deductive powers, but she's proven useful as far as gathering information goes.
whip_the_fools (Ace Attorney) - ☆ ♙
A rather mouthy lawyer who needs to develop more complexity in her insults and less rigidity in her view of the law, in his opinion. For a prosecution prodigy, Sherlock is disappointed that she doesn't seem to have any effect on the state of affairs in the Port. Also, she dissed the Strad. No one disses the Strad.
deathknellgrell (Kuroshitsuji/Black Butler) - ☆☆ ♙
Sherlock definitely didn't expect the torture of demons to involve annoying someone to death. Overall disappointing as a threat and dear lord how he hates their shrieking. More of a pest than anything else.
deadly_jump (Jumper) - ♗
Young British man from a few years before Sherlock's time. Violent and vindictive for reasons yet to be explored. Ella's roommate, so interested in Sionis. He doesn't trust him, though, so he's not letting him into his case. Yet, anyway.
cowboy_newsie (Newsies) - ★★ ♘
Aside from his intriguing psychological profile, Jack is fun to prod because of how easily frustrated he is. As far as Sherlock's concerned, someone with grammar that bad deserves it. Still, Kelly has helped him out several times and has proven more or less dependable. Were he home, Sherlock would definitely have him as one of his London street contacts.
shesmashes (Marvel Comics) - ★★ ♘
One of the two green ladies Sherlock has met in Siren's Port. She-Hulk, however, seems about ten times more stable and hasn't molested him. Yet. A new arrival who actually asked the right sort of questions, Sherlock is just happy to see that she has half a brain stem. Then there's the pressing question of why is she green?
UPDATE: Sherlock is pretty critical of forming a superhero group for Siren's Port; it seems pontificating and unwieldy. This combined with the horrid incident on Valentine's Day has definitely put Shulkie on his irk-o-meter, useful or no. Found out her real name and prefers it, because She-Hulk? Lame.
psyd_seward (Dracula) - ★★ ♘
It's interesting to see how someone from the Victorian Era really acts, though it seems historical accounts were accurate enough after all. At the very least he's a far more interesting conversationalist than half the people he's met at the Port, and it's someone he could actually go on for hours with about turn of the century crime. In the detective's memory banks if he ever needs to profile someone and actually requires help. Rare as that is.
he_is_wild (DC Comics) - ☆☆☆ ♕
It doesn't take someone like Sherlock to figure out this guy is batshit insane. It's difficult to pin down a criminal with no set plan. No morals, he can handle. No method to the madness throws a kink in his logic. Sherlock is biding his time on how to stop this one as Black Mask is more pressing at the moment, and there are plenty of people out for the Joker already.
UPDATE: The Joker has proven time and again that he's one of the most dangerous people in the Port, newcomer or otherwise. The lack of mystery to his actions (i.e. He never keeps a secret about what he's done) is another thing that keeps Sherlock from investigating him, but it's only a matter of time before he or people he's invested in are dragged into the madness.
killswithapen (Death Note) - ☆☆☆☆ ♕
An admitted serial killer, which set off obvious alarms. Whether it was the result of a split personality or not, Sherlock knows those impulses don't just disappear. It doesn't help that his eloquence seems to put him in high regard among some circles. His arrogance almost seems like a challenge to the detective. Definitely on the watch list.
youllsayit (Supernatural) - ☆☆ ♕
The second Satan to show up, and to Sherlock he definitely seems to fit the Devil bill more appropriately than the other. There is a temper, a snakelike coy demeanor, and an obvious sense of superiority to humanity. Sherlock isn't afraid to speak to him, but he's definitely keeping a wary eye on him.
meaculpable (Vertigo/DC Comics) - ★★ ♕
Who knew the Devil would be so relatable to talk to outside of Milton? The title doesn't really impress Sherlock, considering he's not that religious and this Lucifer isn't from his world, but he can hardly deny it fascinating to live among Satan and not be in eternal suffering. His club is far more charming than Hell could ever claim to be, he's quite sure.
makes_asteroids (Marvel Comics) - ★ ♗
Another person Sherlock can stand talking to for more than five minutes, but he definitely senses something deeper than what the older man lets on. Luckily for Magneto, he has his hands too full to investigate that further.
mentis_reae (Ace Attorney) - ☆☆ ♖
It doesn't shock him at all that Edgeworth and von Karma are somehow related. Edgeworth, however, is far more irritating. In some ways, the lawyer reminds him of his brother Mycroft, more in his pomposity than anything else. Even Mycroft could bend a law or two to insure justice gets done. Sherlock does not believe that laws equate with justice, and when Edgeworth seems to stand for that above all else, he can't help but argue with the man until they end up childishly insulting each other. They have similarities, but if you pointed this out to Sherlock, he would probably punch you. In the face. Edgeworth doesn't see things the Sherlock way, so therefore, to Sherlock, he is the biggest idiot in charge at the Port.
tokissity (DC Comics) - ☆☆ ♕
The first green woman he met, which is enough of a draw. As she seems to come from an alternate Earth, he is intrigued to know how she became part plant. And having seen her discuss her own plants with others, he wouldn't mind experimenting on them or getting his hands on some of her poisons. However, this seems to entail her hands on him, and he's not that curious. Definitely keeping an eye on this one, though.
proxysearch (Ergo Proxy) - ★★★ ♖
One of the few officers on the SPPD that actually seems to have an iota of sense to her. It goes a bit far to say that Sherlock likes her, but her methods remind him of Lestrade (i.e., knowing when to take help from someone more keen on doing it). Helped her and others take down the Joker when he began killing police officers. Since the (short lived) arrest of the Joker, Sherlock has worked with Re-l more and more, going to her for police assistance when he cracked the Collar Killer case. He considers her something of a co-conspirator against the Gotham Rogues, and is curious towards her teetering dependency on the law to get work done. They seem to both have become hopelessly entangled with Batman.
UPDATE: Though still a steady police contact, knowing Re-l doesn't come without its ups and downs. When she was accidentally dosed with the new street drug Xaylein, Sherlock and John stopped her from killing herself. Since then, they are two of the few who actually know how to get into Re-l's protected apartment. Sherlock likes to occasionally crash on the couch. And after she attacked him, he knows she's not human. But that's not unusual in the Port, is it?
walkthedawn (Kingdom Hearts) - ★★ ♖
Young man who seems surprisingly on top of things compared to other people in his age bracket in the Port. Currently using his help in uncovering the plans of Master Xehanort and the theft of hearts in the city. An ally by necessity, but at least not an excessively annoying one.
ebony_mask (DC Comics) - ☆☆☆☆ ♕
Why no one else can recognize a sexually perverse, psychopathic and sadistic mob boss when they see one on the NV is quite beyond Sherlock. Roman rubbed Sherlock the wrong way the minute he arrived, and it has since become his pet case to render proof against him, somehow, and keep him from becoming a real public nuisance. Sionis didn't help his cause by shooting Robin; no one breaks Sherlock's toys and gets away with it (Is it secretly decency? Probably). Now knowing he's a murderer that has no qualms in harming children, in addition to following his business contacts, Sherlock's slowly closing in and having a grand old time making Sionis a paranoid bag of nerves in the process.
zealouspeter (Toward the Terra) - ★★ ♘
Now this kid definitely reminds Sherlock of himself at that age. That doesn't stop him from being paranoid about his possible ulterior motives. Definitely a harder enigma to crack than others on the NV.
