Sure! I have a graphic tablet and it makes all easier without doubt! I use the default brush, the one round and hard. If you have the tablet, the only thing to do is this:
Check shape dynamics, set the control on pen pressure et voilà! Depending on the pressure that you'll make, the line will be thinner or thicker. You can also change the minimum diameter to set the min. thickness. If you also want to change the opacity of your strokes, depending on the pressure you make, then check transfer, too. You can set a minimum opacity if you open the tab.
It's pretty hard to do with a mouse, but not impossible; you can simulate the pressure of the graphic tablet using the pen tool. In this case, the brush setting will be this:
