Hm, I don't really have anything specific in mind, but let's see...
Format: Your flash layouts are cool, so I think that coupled with your illustrations could add a neat quality to the layout and set-up. A navigation that's easy to go through is always good too.
Content: I personally love info, so stuff like who were the first women in the industry, and who were the trailblazers and what did they do, I find that interesting. And also about the characters they created or wrote/plotted/drew and what about their contribution that made it special. But having a good balance between the word chunks and visual examples is important too.
Interactivity: Um... I don't know much about this part, I'm not very interactive. ^^;; But I guess voter polls for various content pages would be good, or tying in other interactive sites like Twitter or Facebook. Maybe also allowing input like, 'what/who should be discussed/interviewed next?,' although maybe that falls under the poll category. Could have a forum too, although those are terribly difficult to code so maybe a shoutbox (although those are prone to spam/trolls), or an off-site forum.
Also, you could look around Girl-Wonder.org to get inspiration or ask for help/opinions/interviews -- it's a collection of sites/blogs and community for female comic characters, creators, and fans.
