You put your finger on the one thing I felt was missing this week (everything else I agree was a truly well done, tension-wracked episode) - the brotherly moments felt underplayed somehow. They gave us a little, but I wouldn't have minded more, you know?

Absolutely, I would've really liked a bit more too but with everything that went on this episode I guess I can see why.

But I was a little shocked at the end of the episode that Dean didn't try to go after Sam when he saw the jar of blood. I know it was a "save the world, we're running low on time" situation and all but I really selfishly wanted him to go and find Sam. Is that wrong of me? :P

Nothing - I thought exactly the same thing! I suppose Season 1 Dean would have done just that - I guess it is a mark of maturity of their relationship that he now saves the world before he saves Sam.... or something.

If that's the case, I don't want them to be mature! All I can think of now is Dean at the end of S2 saying "let the world end" after Sam died. :(

When Dean first came across the blood my hope was that Dean would start shouting for Sam, take a couple of steps forward to go look for him and then we'd hear Bobby shouting for Dean and then Dean would remember he had a job to do and regretfully turn back. Silly of me but I would've liked for that to have happened during that little moment. :)

Yes, that would have been perfect - the reluctant tug of duty drawing him away from his first priority, Sam. Ah well, I suppose it was a relatively minor niggle in what was an episode otherwise jam packed with goodness.
